Born 8 September 1814 at Ysgubor Fawr, Chwaen-wen, Anglesey, the eldest son of David Roberts Pughe and Elizabeth his wife. He qualified as a doctor after training at S. Thomas's Hospital, London, where he received the degree of F.R.C.S. He first practised at Barmouth but later moved to Aberdovey where he spent most of his life. In his early days he lived at Clynnog-fawr, Caernarfonshire, and was a close friend of Eben Fardd. In Eben's Cyff Beuno there are many notes by Pughe. He published an entertaining biography of the bard: Eben Fardd: ei nodion a'i hynodion . He also translated Meddygon Myddfai, the Physicians of Myddfai, edited by John Williams, Ab Ithel, and published by the Welsh MSS. Society, 1864. He married Catherine Samuel, daughter of Samuel Samuel, Caernarfon, 21 February 1839; she died 14 September 1862, at Penhelyg, Aberdovey. Four of their sons were physicians, John Eliot Howard (died 1880), Rheinallt Navalaw, Taliesin William Owen (died 1893), who practised at Liverpool, and David Roberts (died 1885), who lived in Montgomeryshire. Their daughter was BUDDUG ANWYLINI PUGHE, the artist, who died in Liverpool, 2 March, 1939, at the age of 83. Buddug Pughe wrote a history of her native district, but the manuscript was never published.

John Pughe was prominent in the public life of Aberdovey and district, being a justice of the peace and a patron of all charitable and religious causes. His religious leanings were towards the Plymouth Brethren, for whom he preached in the small church which he established in the district. He died 9 April 1874, and was buried in the family vault at Cwm Maethlon burial ground.
